Model 410 Flame photometer

The Sherwood Model 410 Flame photometer directly measures Alkali and Alkaline Earth metals
Sodium (Na), Potassium (K), Lithium (Li), Calcium (Ca), Barium (Ba),
Caesium (Cs), Rubidium (Rb) and Strontium ( Sr) by means of a low temperature
flame using propane, butane or Natural gas. The measurement on the Model
410 Clinical Flame Photometer can be semi automated by means of the
Model 805 Clinical Diluter and for clinical samples the ease of use
is enhanced by the Lineariser Option which enables results in the clinically
significant concentrations to be displayed directly on the Model 410
Clinical Flame Photometer in mmol/l without the need to prepare a standard
curve. The Model 805 Clinical Diluter is ideal for diluting such samples
and the dilution ratios are 200:1 for serum Na and K and 50:1 for Lithium
The principles of flame emission spectroscopy were first investigated
by Kirchoff and Bunsen (1860s). In the 1920s Lundengardh refined the
technique by incorporating a nebuliser and electro optical detector
which paved the way to modern quantitative analysis.
Our predecessor company Evans ElectroSelenium Ltd (EEL) developed
the first Model 100 Flamephotometer

<>
<>The Model 360 Flame
Photometer's mixing chamber, burner stem and burner head are taken from
our world renowned Model 410 Flame Photometer. Automatic flame optimisation
comes from our Dual Channel Flame Photometer Model 420 series of instruments.
Put simply, the Model 360 Flame Photometer is a new product built
with tried and tested components where it really matters. It has the
best limits of detection available from a low cost instrument and precision
comparable to higher cost instruments.
The five element filter stick is easy to remove for cleaning and
filter replacement. The mixing chamber, burner head and burner stem
come apart for cleaning with no tools required. Electronics and pneumatics
come in separate "pods" and both are easily removed from the main instrument
chassis; offering a potentially huge saving on shipping costs if a return
to base repair or replacement becomes necessary.
The appropriate filter for the element to be analysed is selected
using the filter stick and indicated by LEDs on the front panel, which
also displays flame status by LED. A display "hold" button allows readings
to be captured until recorded - useful if you are unexpectedly called
away. For ease of operation, the air regulator is accessible and pressure
gauge visible at the front.
A large sample work area is also provided in a spill containment
tray which is easy to remove and clean. The instrument is supplied ready
for use following installation of the chimney. Connection to the mains
power supply is easy; each unit is supplied with a universal switch
mode power supply with its own multi-plug adapter kit; no need to worry
about voltage selection or making up power cords

MODEL 926 CHLORIDE ANALYSER

The Sherwood Model 926 is used for the determination of chloride
ions. It is an instrumental analogue of "Argentimetry", the traditional
titrimetric methods using silver nitrate reagent. Like these classic
methods it relies on the chemical formation of the very insoluble salt,
silver chloride.
The importance of chloride determination has been realized for well
over a century, with many variations and changes being made to the techniques
in order to improve the detectability and selectivity. Research into
the analysis of chloride was conducted by Gay-Lussac (1832), Levol (1853),
Mohr (1856) and Volhard (1874) and their findings have proven to be
the basis of the methods which are still in common use today.

Magnetic Susceptibility is defined as:
"The ratio of the intensity of magnetism induced in a substance to
the magnetising force or intensity of field to which it is subject"
Basic Principles of Magnetic behaviour
Based on their magnetic properties, all substances can be classified
into one of three groups, those attracted by a strong magnetic field,
known as paramagnetic, those repelled, designated diamagnetic, and,
finally, the most recognised class, ferromagnetic, unique in their ability
to retain their own magnetic field. Ferromagnetics are able to retain
a permanent magnetic field since their free electrons are in close proximity
and remain aligned even after the external magnetic field is removed.
Unlike the ferromagnets, the magnetic properties of the diamagnetic
or paramagnetic materials could only be observed and measured when these
samples are held within a magnetic field applied externally

The Model
260 Programmable Colorimeter

The basic design of the
Sherwood CHROMA Colorimeter has been stable for several years and they
have an enviable world-wide reputation for rugged performance. For the
21st century Sherwood Scientific has reduced the range, updated its
Photometers and added a software package unique in this class of instrument.
The Model 260 Colorimeter offers features not elsewhere available and
provides solutions to the requirements of quality control, water testing
and for the clinical chemist for low sample runs.
The Model 260 can be battery powered and thus can be used outside
the Laboratory.
The CHROMA Model 260 Programmable Colorimeter
This Model M260 replaces the CHROMA 257 Colorimeter
and incorporates full programmability allowing up to 100 methods to
be installed by the user. This permits semi-automatic operation with
the user only required to insert the prompted filter. An RS232 output
allows the use of a serial printer and every result, blank and calibration
is time stamped. The unit allows quick Absorbance and Transmission measurements
with Auto-Zero, Concentration with Standard Solutions or Factor as well
as the new Kinetic Reaction Rate and End Point methods.
Temperature and the new CHROMA range
The Model 260 Colorimeter has two options to control the temperature
of reactions. The low cost option is to use external temperature control
by means of solid block heating. The software built in to all three
of the new models includes the setting of Incubation (or lag time) and
Measuring time. Towards the end of these times the user is alerted to
return the cuvettes containing the blank sample or calibrant to the
colorimeter by means of a buzzer. The relevant reading is taken and
the cuvette is returned to the heating block to complete the rest of
the test.
Sherwood has also developed an electrically heated cuvette holder
(267 86 001) designed specifically for the micro cuvettes (001 26 120).
This is able to control the temperature to 37 C plus or minus 0.1 degrees
C. and is available as an option.

WHAT IS FLUID BED DRYING?
By forcing enough air through a bed of particles, the particle bed may
assume a fluid-like state (resembling a boiling liquid). Heating the
incoming air and managing air flow rate through the sample, the Model
501 Fluid Bed Dryer provides thorough mixing and maximum contact of
solid with moving air.
The result; a process producing more even and much quicker drying
than conventional methods
FAST
Delivering up to 2.5m3 per minute of air, the Model 501 Fluid
Bed Dryer can break up wet samples and ensure vigorous mixing and rapid
moisture removal.
5kg of wet "ideal" sample (80% moisture) can be dried in 15-20 minutes
(5 Litre tub).
NON-AGGLOMERATING
Air separated particles prevents lumps and caking, both of which make
other drying processes much slower.MILD High air flow rate gives:
-
high moisture removal rates at relatively low temperatures
-
thorough mixing, so no wet spots requiring extra thermal energy
to penetrate
-
an air cushion between particles to reduce abrasion and particle
size alteration
HOMOGENEITY OF SAMPLE
Static drying methods leave evaporation residues at the sample surface
giving a heterogeneous sample. Fluid Bed Drying achieves the opposite.
Mixing during Fluid Bed Drying gives homogeneous samples, making an
ideal method of representative sample preparation for subsequent material
analyses
REPRODUCIBLE
Microprocessor control of air flow, inlet air temperature and drying
period coupled with fluid bed action gives highly reproducible experiments
and finished samples. After preliminary experiments, a known moisture
content in the final sample (ideal for tablet forming) or removal of
external (surface) moisture only, may be achieved. Drying times to required
moisture content may be optimised and drying patterns studied to aid
scale-up and plant design.
Without a PC connected the Model 501 Fluid Bed Dryer can run one stored
program of up to 16 steps (previously downloaded from a PC), or may
be used as a conventional (manually controlled) Fluid Bed Dryer. Additional
features of this advanced in-lab dryer technology include:
- Precise air flow feed-back control
- Membrane sealed controls to prevent ingress of particles into
the instrument
- Reduced operating noise

Cellular Antigen Stabilising Reagent
Application - TransFix®
Flow Cytometry comparison of fresh whole blood
with a TransFix®
TransFix® Fresh Blood Samples Day 0 --- (No TransFix®)

TransFix® Blood Sample Day 10

Flow Cytometry comparison of fresh whole blood
with a TransFix® stabilised sample at Day10 demonstrates
that TransFix® stabilised whole blood has the same flow
cytometry staining pattern as seen in fresh whole blood. Staining
examples with (CD3 FITC and CD4 APC) and (CD3 FITC and CD8 PE) antibodies
are shown above.
Immunophenotyping
Subsets of leucocytes can be distinguished on the basis of cell surface
antigens using flow cytometry. Quantitative and qualitative changes
in leucocyte sub sets are used to identify and monitor haematologic
diseases. TransFix® has been shown to preserve a wide range
of cellular markers (Barnett et al, 1999) for up to 10 days.
A full haematological profile can also be obtained up to 7 days after
the addition of TransFix®.
The wide range of clones and conjugates available make it essential
that all conjugates are tested in conjunction with TransFix®
to ensure there is no interference with antibody binding. Care should
also be taken with regards to intracellular antigens as TransFix®
causes a progressive permeabilisation of the cellular membrane. As a
direct result intracellular markers will leach out over time. As a general
guide intracellular antigen measurement on TransFix® treated
samples is only possible up to 3 days after the addition of TransFix®.
Other Species
TransFix® has been tested on mouse, rat, guinea pig, sheep,
horse, pig and green turtle blood samples. The amount of TransFix®
required to stabilise blood varies considerably in different species.
In some species the volume required also varies by breed and type.
The user must determine the optimal dilution required. The optimal
dilution is the concentration where the same flow cytometry light scatter
result remains unchanged after the treated sample has been stored for
10 days at 4 degrees C.The protocol for
determining the optimal concentration needed can be downloaded here.
Functional Assays
Samples treated with TransFix® cannot be used in functional
assays.
Storage
TransFix® is stored at room
temperature (18 - 24<
>
degrees C)
CytoFix®is a cellular antigen stabilisation procedure
that ensures the retention of leucocyte light scatter and immunological
staining characteristics of stabilised whole blood and buffy coat for

C). In countries where the ambient temperature is greater than 24
degrees C TransFix® can be stored at 4 degrees C. However,
it must be allowed to warm up to room temperature before use. TransFix®
must NEVER be frozen.
Wherever possible TransFix® treated samples should be
refrigerated at 4<> degrees <>C. Treated samples
are also stable at temperatures up to 25<> degrees
C for 7 days and at temperatures between 26<>
degrees C and 37<> degrees <>C for 4
days.
History
TransFix® was developed by medical scientists from UK
NEQAS, which is part of Sheffield Teaching Hospitals NHS Foundation
Trust. TransFix® was developed out of the requirement to
produce stabilised blood samples that could be used in External Quality
Assurance programs for flow cytometry. The first publication on TransFix®
as a cellular antigen stabilizing reagent was made in 1999 by D. Barnett
et al.
TransFix® has been in circulation in the research world
from 1999 until the present day, generating a number of scientific publications.
In 2005 the Sheffield Teaching Hospitals NHS Foundation Trust licensed
TransFix® to Cytomark in order to make it more generally
available to the clinical and scientific world.

Back ground . . .
A potentiostat is a
control and
measuring device. It comprises an
electric circuit which controls the potential across the cell by
sensing changes in its
resistance, varying accordingly the current supplied to the
system: a higher resistance will result in a decreased current,
while a lower resistance will result in an increased current, in
order to keep the voltage constant as described by
Ohm's law.
-

As a result, the variable system
resistance and the controlled current are
inversely proportional

is the output electrical current of the potentiostat
- Ec

The
Impedance Spectroscopy
technique
greatly expands the application range of electrochemical measures and
helps in defining the dynamic behaviour of reactions as well as identifying
the relative contribution of two or more concurrent processes. So far,
the premium to be paid to enter this arena was a significant amount
of money and, sometimes, the need to change the whole measurement chain.
The new 520requency Response
anlyser
from Materials Mates Italia sets a new level of performance and flexibility
at a price that makes its selection painless. Covering a wide frequency
range (10 μHz - 2 MHz)
with great accuracy (up to 0.1% amplitude, 0.05deg phase)
the interfaces with any potentiostat
with analog I/O connections. ZedScope,
its dedicated software package, offers all the
features needed for the most difficult tasks with an intuitive approach.
Thanks to its architecture, the
520 will accept further software packages for pulsed
techniques, soon available from Materials Mates Italia. Upgrade now your equipment

Dielectric spectroscopy (sometimes called impedance spectroscopy), and also known as electrochemical impedance spectroscopy, measures the dielectric properties of a medium as a function of frequency. It is based on the interaction of an external field with the electric dipole moment of the sample, often expressed by permittivity.
It is also an experimental method of characterizing electrochemical systems. This technique measures the impedance of a system over a range of frequencies, and therefore the frequency response of the system, including the energy storage and dissipation properties, is revealed. Often, data obtained by EIS is expressed graphically in a Bode plot or a Nyquist plot.
Impedance is the opposition to the flow of alternating current (AC) in a complex system. A passive complex electrical system comprises both energy dissipater (resistor) and energy storage (capacitor) elements. If the system is purely resistive, then the opposition to AC or direct current (DC) is simply resistance.
Almost any physico-chemical system, such as electrochemical cells, mass-beam oscillators, and even biological tissue possesses energy storage and dissipation properties. EIS examines them.
